What is a 3X4 genetic test — and why do I use it?

Not all genetic tests are created equal — and if you’ve ever wondered what actually goes into the testing I use in my practice, this is for you.

The 3X4 Genetic Blueprint is a simple cheek swab, done once. It analyses 157 carefully selected genetic variants (SNPs), each chosen because it has a proven functional effect in clinical practice — meaning it can be influenced through diet, lifestyle and targeted supplementation.

Many genetic tests look at thousands of SNPs. More data sounds better, but in practice it rarely is.

There’s no point knowing about a gene if there’s nothing we can do clinically to change the outcome.

We’re not looking at genes to diagnose disease. We’re looking at where your susceptibilities lie, which pathways need support, and what we can actually do about them.

Pathways, not just genes

Most genetic reports give you a list of individual gene variants and leave you to figure out what they mean together. 3X4 organises results by pathway instead.

So rather than seeing “MTHFR — variant detected” in isolation, you see the entire methylation pathway — every gene involved, and how the pathway as a whole is functioning.

It’s the difference between knowing one instrument is out of tune and hearing how the whole orchestra sounds.

What my own results showed

When I got my own results back, I was genuinely surprised by what came up.

I have a lot of variants in the methylation cycle, so I expected methylation to be my top pathway. Instead, inflammation ranked even higher — which I hadn’t seen coming.

In the systems pathways, collagen and joints, bone health, and histamine overload all came back very high. Histamine was no surprise — and it helped explain the inflammation result. But bone health and collagen and joints? That hadn’t been flagged by any other testing I’d done previously.

And then when I thought about it — I have a family history of osteoporosis. Suddenly it made a lot more sense, and it put a bone density (DEXA) scan at the top of my to-do list.

One result I found genuinely interesting was my variant on the CLOCK gene, which helps regulate your circadian rhythm. It made a lot of sense — I’ve always been a night owl, and you will never see me at the 5am club!

That’s the kind of insight that changes how you approach your health — not because it’s alarming, but because it’s specific and actionable.

What the report covers

The report organises your results across 10 categories, covering 35 metabolic pathways in total:

  • Cellular — methylation, inflammation, oxidative stress, detoxification

  • Brain — memory, mood & behaviour

  • Heart & Vascular — vascular health, blood pressure, blood clotting, cholesterol

  • Weight — pro-inflammatory fat, fat storage, energy expenditure, appetite

  • Hormones — hormone balance

  • Blood Sugar Regulation — glucose & insulin

  • Muscle & Skeletal — collagen & joints, bone health, muscle health

  • Activity — injury, endurance, recovery, power, training response

  • Sensitivities — salt, histamine overload, caffeine, gluten, iron overload

  • Nutrients — vitamin B12, choline, folate, vitamin D, fatty acids, vitamin C

Each pathway is colour-coded from dark purple (highest impact) to light green (lowest impact) and ranked, so you can see at a glance where to focus first. The report also shows your protective steps — genetic factors that are working in your favour — so you get a balanced picture, not just a list of things to fix.

It’s all delivered through the 3X4 Health app, with targeted recommendations for each pathway. You still get the individual SNP detail if you want it (a practitioner favourite!).

What the test looks like

Here’s a quick look at how the test is done — it’s as simple as a cheek swab at home.

One test, for life

Your genetics don’t change — test once, and you have a roadmap for life. It shows you what to build your health around, from food and movement to sleep and which supplements are likely to suit you, and becomes one piece of the bigger picture as your health evolves.

The 3X4 genetic test is a core part of the Ageless Foundation Program, which combines your genetic results with comprehensive blood testing and personalised nutrition guidance.
